WHIRLY2 is a nucleus-encoded, plant-specific nucleic-acid-binding protein that localizes predominantly to mitochondria, where it is enriched in mitochondrial nucleoids and associates with mtDNA-containing complexes. It is synthesized with an N-terminal organelle-targeting presequence that is removed upon import, yielding a mature protein whose core is the conserved WHIRLY domain, an ssDNA-binding module typified by the KGKAAL motif; WHIRLY-family structural studies indicate this domain supports oligomerization into tetramers that can further assemble into higher-order particles on long ssDNA. Functionally, WHIRLY2 acts as an architectural nucleoid factor that binds ssDNA largely without strong sequence preference to promote mtDNA organization/compaction and to support mitochondrial genome stability; genetic disruption in Arabidopsis leads to disordered nucleoids and altered mitochondrial ultrastructure (including reduced cristae organization) accompanied by impaired mitochondrial performance, and loss-of-function also compromises mtDNA integrity/repair under stress (e.g., salt stress) with accumulation of aberrant recombination products and heightened stress sensitivity. Collectively, WHIRLY2 links nucleoid structure to mitochondrial morphology and bioenergetic competence, helping maintain mitochondrial genome maintenance and stress-responsive mitochondrial function.
