          ZNRF3 Antibody (PA5-143891) in ICC/IF

          Immunocytochemistry analysis of ZNRF3 in U2OS cells. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent for 15 mins. The cells were blocked with 10% goat serum. Samples were then incubated in ZNRF3 Polyclonal antibody (Product # PA5-143891) using a dilution of 5 μg/mL. DyLight®488 Conjugated Goat Anti-Rabbit IgG was used as secondary antibody at 1:100 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI.           Product Specific Information

          Adding 0.2 mL of distilled water will yield a concentration of 500 µg/mL.

          Positive Control - WB: human CACO-2 whole cell, human A549 whole cell, human Hacat whole cell. ICC/IF: U20S cell. Flow: PC-3 cell.|Store at -20°C for one year from date of receipt. After reconstitution, at 4°C for one month. It can also be aliquotted and stored frozen at -20°C for six months. Avoid repeated freeze-thaw cycles.

          Target Information

          The ZNRF3 gene encodes a transmembrane E3 ubiquitin ligase that plays a critical role in regulating the Wnt signaling pathway, which is essential for various biological processes, including embryonic development and cell proliferation. ZNRF3 functions by negatively regulating this pathway through ubiquitination and degradation of Frizzled receptors, thereby suppressing the Wnt signaling. Located on chromosome 22, ZNRF3 acts as a tumor suppressor gene by inhibiting oncogenic Wnt signaling and has been implicated in the formation and progression of several cancers, particularly colorectal cancer. Moreover, ZNRF3 interacts with R-spondin proteins which are known to augment Wnt signaling by targeting ZNRF3 for degradation, thus facilitating cell differentiation and proliferation. Alterations in ZNRF3 expression or mutations can lead to aberrant Wnt signaling, contributing to tumorigenesis. Additionally, ZNRF3's role extends to its participation in lipid metabolism and maintenance of bone homeostasis. It has been studied as a potential therapeutic target due to its central involvement in these regulatory pathways.
