The 1,25 Dihydroxyvitamin D3 (DHVD3) ELISA quantitates DHVD3 in serum, plasma and other biological fluids.
Principle of the method
The Competitive mulispecies ELISA research use only kit is designed to quantitatively measure 1,25 Dihydroxyvitamin D3independent of species . A 1,25 Dihydroxyvitamin D3 standard is provided to generate a standard curve for the assay and all samples are read off a user generated standard curve.Standards or diluted samples are pipetted into a coated microtiter plate and Biotinylated Detection Ab specific to 1,25 Dihydroxyvitamin D3 is added. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each well and incubated. Then a TMB substrate solution is added to each well. The enzyme substrate reaction is terminated by the addition of stop solution and the color change is measured on a microplate reader. The concentration of 1,25 Dihydroxyvitamin D3 in the samples is then determined by comparing the OD of the samples to the standard curve.

1,25-dihydroxy vitamin D3, or calcitriol, is the active form of vitamin D3. It functions as a hormone and plays a crucial role in regulating calcium and phosphorus metabolism. It is mainly localized in the intestines, bones, kidneys, and parathyroid glands. Structurally, it is a steroid hormone derived from cholesterol. Its functions include promoting calcium and phosphorus absorption, maintaining bone health, regulating blood calcium levels, and suppressing parathyroid hormone production.
