The mouse Interleukin-10 (IL-10) ELISA kit is designed for the quantitative measurement of mouse IL 10 in a range of biological samples, including murine serum, plasma, buffered solutions, or cell culture medium. This assay has high specificity and can accurately detect both natural and recombinant forms of mouse IL-10.
Principle of the method
The mouse IL-10 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) employs a well-matched pair of antibodies to capture and quantify the target analyte. The microplate wells are coated with a specific antibody that selectively binds to Hu IL-10. Subsequently, the samples, standards, or controls are added to the wells, allowing the target to bind to the immobilized (capture) antibody. The addition of a second (detector) antibody forms a sandwich complex. A substrate solution is then introduced, initiating a reaction with the enzyme-antibody-target complex, leading to the generation of a measurable signal. The intensity of this signal is directly proportional to the concentration of Hu IL-10 present in the original specimen.
IL-10 encodes a protein that acts as a cytokine and is primarily produced by monocytes, with some production by lymphocytes. This cytokine has various effects on immunoregulation and inflammation, including down-regulating the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. Additionally, IL-10 can block NF-kappa B activity and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice have suggested that this cytokine is an essential immunoregulator in the intestinal tract. Mutations in this gene have been associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis. Diseases associated with IL10 include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1.
