About This Kit

The multispecies Prostaglandin E2 (PGE2) ELISA quantitates PGE2 in serum, plasma, and other biological fluids

Principle of the method:

The Competitive multispecies ELISA research-use-only kit is designed to quantitatively measure Prostaglandin E2 (PGE2) independent of species. A PGE2 standard is provided to generate a standard curve for the assay and all samples are read off a user-generated standard curve. Standards or diluted samples are pipetted into a coated microtiter plate and Biotinylated Detection Ab specific to PGE2 is added. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color change is measured on a microplate reader. The concentration of PGE2 in the samples is then determined by comparing the OD of the samples to the standard curve.

The PTGES2 gene encodes for the enzyme prostaglandin E synthase 2 (PTGES2), which is crucial for the biosynthesis of prostaglandin E2 (PGE2) from arachidonic acid. This enzyme operates downstream of the cyclooxygenase (COX) pathway, particularly interacting with COX-1 and COX-2 enzymes. PTGES2 is involved in various physiological and pathological processes by generating PGE2, which functions in inflammation, fever generation, and pain. Notably, PTGES2 is a membrane-associated protein and is distinct in its role compared to other prostaglandin synthases due to its specific functional and regulatory properties. Its activity is essential in fine-tuning the levels of PGE2 in tissues, impacting numerous signaling cascades, including those involved in immune responses and vascular functions.
