The multispecies Reverse Triiodothyronine (rT3) ELISA quantitates rT3 in serum, plasma, and other biological fluids
Principle of the method:
The Competitive multispecies ELISA research-use-only kit is designed to quantitatively measure Reverse Triiodothyronine (rT3) independent of species. A rT3 standard is provided to generate a standard curve for the assay and all samples are read off a user-generated standard curve. Standards or diluted samples are pipetted into a coated microtiter plate and Biotinylated Detection Ab specific to rT3 added. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color change is measured on a microplate reader. The concentration of rT3 in the samples is then determined by comparing the OD of the samples to the standard curve.

Reverse triiodothyronine (rT3) is a biologically inactive form of the thyroid hormone triiodothyronine (T3). rT3 is produced by the enzymatic deiodination of T4, primarily in peripheral tissues, and is mainly found in the liver and kidneys. Its primary function is to regulate the metabolism of T3 by blocking its action. The structure of rT3 is similar to that of T3, with the exception of the position of one iodine atom. This structural difference causes rT3 to be biologically inactive and unable to bind to thyroid hormone receptors. Despite its lack of direct physiological activity, rT3 levels can be used as a marker of thyroid hormone status and metabolic function.
