Basophil Marker Antibody (53-9748-42) in Flow
Normal human peripheral blood cells were surface stained with Anti-Human CD123 APC (Product # 17-1239). The cells were then permeabilized with the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824), and intracellularly stained with Mouse IgG1 K Isotype Control Alexa Fluor® 488 (Product # 53-4714) (left) or Anti-Human Basophil Marker Alexa Fluor® 488 (right). Cells in the lymphocyte gate were used for analysis.

Product Specific Information
Description: The 2D7 monoclonal antibody recognizes human 2D7 antigen expressed exclusively by human basophils. This human basophil marker is a 7.2-7.5 kDa protein present exclusively in the secretory granules of human basophils. Upon activation, basophils may degranulate, thereby decreasing the amount of 2D7 antigen available for staining. This antibody does not stain mast cells.
Applications Reported:This 2D7 antibody has been reported for use in intracellular staining followed by flow cytometric analysis.
Applications Tested: This 2D7 antibody has been pre-titrated and tested by intracellular staining and flow cytometric analysis of normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (cat. 88-8824) and protocol. Please refer to BestProtocols®: Protocol A: Two step protocol for (cytoplasmic) intracellular proteins located under the Resources Tab online. This antibody can be used at 5 µL (0.5 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.
Excitation: 488 nm; Emission: 519 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.
