The Thermo Scientific Pierce Cleaved PARP Colorimetric In-Cell ELISA Kit is a near infrared microplate assay optimized to measure activated (cleaved) poly (ADP-ribose) polymerase (PARP) and tubulin in whole cells.

Features of the Cleaved PARP Colorimetric In-Cell ELISA Kit:

• Measures activated (cleaved) poly (ADP-ribose) polymerase (PARP) and tubulin in whole cells using 96-well or 384-well microplates
• Colorimetric detection kit for use with standard microplate readers
• Whole-cell detection requires no cell lysis, protein extraction or special sample preparation
• Provides better data and higher throughput than quantitative western blotting
• Less costly (one well vs. one lane) than western blotting

This ELISA kit for whole cells includes a rabbit monoclonal anti-(cleaved) PARP antibody, mouse monoclonal anti-alpha-Tubulin antibody, detection reagents and all buffers required for colorimetric detection. The kit enables simultaneous detection of both Cleaved PARP and Tubulin as a reference.

Overview of PARP:
Poly (ADP-ribose) polymerase (PARP) plays a role in repair of single stranded DNA nicks. PARP also regulates proteins such as histones and telomerase through ADP-ribosylation and is an important marker of caspase 3-mediated apoptosis. In response to apoptotic signals, activated caspase 3 cleaves the 116 kDa PARP protein into 85 kDa and 25 kDa fragments. In humans, PARP cleavage occurs at Asp 214 and Gly 215 residues. Cleavage of PARP correlates with DNA fragmentation and other morphological changes making it a critical marker for apoptosis. The Cleaved PARP In-Cell ELISA Kit enables simultaneous detection of both Cleaved PARP and Tubulin as a reference.
