Applied Biosystems TaqPath DuraPlex 1-Step RT-qPCR Master Mix is a benchtop-stable, single-tube master mix optimized for rapid, sensitive, and reproducible detection of viral and bacterial pathogens even in the presence of PCR inhibitors. This automation-friendly master mix is ideally suited for testing and developing high-throughput molecular diagnostics, as well as for gene expression analysis for high-throughput biologics screening. The 4X concentrated formulation can multiplex six targets in a single well and has intuitive user handling.
TaqPath DuraPlex 1-Step RT-qPCR Master Mix is available in two formulations:
• With ROX dye as a passive reference—recommended for multiplex assays with up to five targets in a single well*
• Without ROX dye as a passive reference—recommended for multiplex assays with up to six targets in a single well
*Not recommended for use with assay utilizing JUN (or similar emission wavelength) dye.

Support workflow automation
TaqPath DuraPlex 1-Step RT-qPCR Master Mix enables a full workday (over eight hours) of stability of completely assembled reactions (master mix, sample, and assay), helping enable the use of liquid handling and other automation equipment. This fast-cycling master mix can be used effectively for both RNA and DNA detection of bacterial, viral, and fungal targets. The figure below shows consistent sensitivity with a multiplex assay tested at 0 and 8 hours compared to other manufacturers.
Optimized for high-order multiplexing
TaqPat DuraPlex 1-Step RT-qPCR Master Mix has been validated with multiplex assays with up to six targets in a single well with the No ROX formulation and up to five targets in a single well with the ROX formulation. It enables accurate detection of both low and high RNA and DNA concentration samples and provides wide dynamic range. The figure below demonstrates the excellent PCR linearity over an 8-log input range in a 5-plex assay.
Improved handling
TaqPath DuraPlex 1-Step RT-qPCR Master Mix has been formulated to improve in-use handling, including a lower viscosity to allow for more effective pipetting, including with automated liquid-handling platforms. The master mix is stable at room temperature (24°C) for up to 1 week and can be stored under refrigeration (4°C) for up to one month after first use It can be used confidently through eight freeze-thaw cycles without loss of performance. The novel formulation helps ensure that the master mix can be adapted to a variety of unique workflow scenarios. The figure below shows consistent performance of reagents stored for one week at 24°C .
Increase confidence
The unique proprietary TaqPath DuraPlex 1-Step RT-qPCR formulation allows robust performance even in the presence of substances that can inhibit PCR, resulting in effective template detection from both purified RNA and DNA, including challenging clinical sample matrices such as VTM, stool, blood, and urine. The figure below shows performance with common PCR inhibitors compared to master mixes from other manufacturers.
Facilitate reproducibility
This General Purpose Reagent (GPR), labeled 'For Laboratory Use', is manufactured in an ISO 13485-certified and FDA-registered facility that adheres to the principles of current good manufacturing practices (cGMP). All lots of TaqPath DuraPlex 1-Step RT-qPCR Master Mix are functionally tested to help ensure lot-to-lot reproducibility for Ct and dynamic range consistency across a wide variety of assays. The figure below shows consistent performance of three lots of master mix.
