Thrombopoietin Antibody (MA5-57973) in WB
Western blot analysis of Thrombopoietin in Recombinant Mouse Thrombopoietin/Tpo Protein lysates (5 and 10 ng/lane). Samples were incubated overnight at 4°C with Thrombopoietin Recombinant Monoclonal Antibody (Product # MA5-57974) using a dilution of 1:1, 000. Followed by a HRP-conjugated Goat anti-Rabbit IgG (H+L) Secondary Antibody at 1:10,000 dilution. Blocking buffer: 3% nonfat dry milk in TBST; Exposure: 20 seconds.

Target Information
Thrombopoietin (TPO, THPO, MPL ligand, megakaryocyte colony-stimulating factor) is responsible for the generation of platelets through its action on hematopoietic cells committed to the megakaryocyte lineage. Binding of thrombopoietin to its receptor, MPL (CD110), initiates a signaling cascade via JAK/STAT, MAPK, and PI3K/AKT signaling pathways that leads to the proliferation and differentiation of megakaryocytes and, ultimately, to the production of platelets (thrombopoiesis). Thrombopoietin is constitutively produced by the liver and has a predicted molecular weight of 38 kDa.
