          PBRM1 Antibody (PA5-84414) in ICC/IF

Immunofluorescence analysis of PBRM1 was performed using 70% confluent log phase HEK-293 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The cells were labeled with PBRM1 Polyclonal Antibody (Product # PA5-84414) at 1:100 dilution in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A... Target Information

The PBRM1 gene encodes a subunit of the PBAF (Polybromo-associated BAF) chromatin remodeling complex, playing a crucial role in the regulation of chromatin structure and transcription. PBRM1, also known as BAF180, contains bromodomains that recognize acetylated lysine residues on histone tails, facilitating chromatin remodeling necessary for effective transcriptional regulation. This gene is frequently mutated in renal cell carcinoma, particularly clear cell renal cell carcinoma (ccRCC), where such mutations are implicated in the pathogenesis of the disease. PBRM1 acts as a tumor suppressor, and its loss of function can lead to deregulation of gene expression, contributing to oncogenic processes. It is involved in pathways that govern cell cycle regulation and DNA damage response, reflecting its importance in maintaining genomic stability. Ongoing research focuses on understanding PBRM1's interactions within chromatin remodeling complexes and its potential as a therapeutic target in cancer treatment.
