The SS18/SSX fusion gene is pivotal in the pathogenesis of synovial sarcoma, a rare type of cancer typically arising in the soft tissues around joints in adolescents and young adults. The SS18-SSX fusion results from an exchange between chromosome 18's SS18 gene and one of the SSX genes (often SSX1 or SSX2 on the X chromosome), creating a hybrid protein responsible for oncogenic transformation. This fusion protein interacts with the SWI/SNF (BAF) chromatin remodeling complex, disrupting normal gene regulation and leading to tumorigenesis. Clinically, the SS18/SSX fusion serves as a crucial molecular diagnostic marker, aiding in the identification of synovial sarcoma. Moreover, SS18-SSX fusion proteins lack DNA-binding motifs but still effectively alter gene expression through epigenetic mechanisms. Despite the establishment of this fusion gene as a diagnostic marker, its prognostic value concerning survival rates remains debated.
