The Ion GeneStudio™ S5 System is a semiconductor-based next-generation sequencing (NGS) system that enables simple, flexible targeted sequencing workflows for clinical research and research laboratories. From targeted gene panels to exomes and transcriptomes, the Ion GeneStudio S5 system helps laboratories manage both small and medium-throughput sequencing projects across oncology, inherited disease, reproductive health, infectious disease, microbial genomics, and transcriptomics applications.

With cartridge-based reagents and the flexible Ion 4-Series chip portfolio, the system is simple to use while providing scalable sequencing throughput. The Ion GeneStudio S5 system supports a broad range of targeted DNA sequencing and RNA sequencing applications, enabling laboratories to increase sample throughput or sequencing depth using the same proven workflow.

The Ion GeneStudio™ S5 system is the base model within the GeneStudio family that also includes the GeneStudio S5 and the Ion GeneStudio S5 Prime systems. It supports the 510 up to the 540 Chips, but it cannot run the largest 550 and 560 Chips, compatible only with the GeneStudio Plus or Prime.

The Ion GeneStudio™ S5 Series

The Ion GeneStudio™ S5 systems share the same foundational semiconductor sequencing technology and fluidic architecture across all three instruments: the Ion GeneStudio™ S5 system, the Ion GeneStudio™ S5 Plus system, and the Ion GeneStudio™ S5 Prime system. The primary difference across the three instruments is on-sequencer analysis speed and supported chips. While the GeneStudio™ S5 Plus and Prime systems can run any of the Chips, from the 510 to the 560, the GeneStudio™ base model can only support the 510, 520, 530 and 540 Chips.

The Ion GeneStudio S5 Series leverages the speed of semiconductor sequencing to produce high-quality sequencing data in just a few hours. When paired with the Ion Chef™ System, laboratories can go from library preparation to sequencing data in as little as 24 hours with approximately 45 minutes of total hands-on time.

With single-day installation, a straightforward user interface, and cartridge-based reagents, the Ion GeneStudio S5 systems are easy to use benchtop sequencers. Combined with Ion AmpliSeq™ technology for targeted panel design, the Ion Chef™ System for automated library and template preparation, and Ion Reporter™ Software for automated variant annotation, targeted NGS workflows are simpler than ever.

Small sample input

Low-input DNA and RNA from challenging sample types—including formalin-fixed paraffin-embedded (FFPE) tissue, fine needle aspirates, and circulating cell-free DNA and RNA (cfDNA/cfRNA)—can be difficult to sequence on other next-generation sequencing platforms. With Ion AmpliSeq technology and the Ion GeneStudio S5 Series, laboratories can sequence with as little as 1 ng of DNA or RNA input.

The Ion Sphere Particles (ISPs) used depend on the chip type selected for the workflow. Each Ion Chef kit includes ISPs that are specific to the corresponding chip type to support template preparation.

What is the difference between the Ion S5 System and the Ion GeneStudio S5 System?

The Ion S5 System and the Ion GeneStudio S5 System have the same functionality and support the same consumables, chip types, and assays. The Ion S5 System was rebranded as the Ion GeneStudio S5 System, and only the GeneStudio S5 System is currently manufactured.

Can I run the Ion GeneStudio S5 System/Ion Chef Instrument at lower humidity levels (10-20% humidity) rather than the 40% humidity recommended in the user guide?

We do not recommend running the Ion GeneStudio S5 System/Ion Chef Instrument in environmental conditions other than our recommended operating conditions. When the instrument runs within a lower humidity range of 10-20% there are risks to the electronics, as low humidity levels are likely to lead to electrostatic discharges. Damage to the electronics can result in run failure and hence performance cannot be guaranteed. Please ensure that the instrument is run at the recommended humidity of 40%.
