Thermo Scientific SMART Digest Dual Enzyme columns containing pepsin and protease XIII provide fast, reliable digestion of proteins for for HDX-MS analysis helping to provide the sequence characterization of biotherapeutics.

SMART Digest Dual Enzyme columns for HDX-MS analysis help to provide the characterization of biotherapeutics.

The combination of enzymes are complementary, creating many short, overlapping N-terminal peptides for a high degree of sequence coverage.

Pepsin cleaves proteins preferentially at carboxylic groups of aromatic amino acids such as phenylalanine, tryptophan and tyrosine. It will not cleave at bonds containing valine, alanine or glycine. The optimum pH range for pepsin activity is 1.0–3.0.

Proteinase XIII cleaves proteins preferentially at carboxylic groups of basic amino acids (arginine, lysine, histidine). Like pepsin, Proteinase XIII is active at low pH.
