L’isomère simple, 6-FAM, contient un acide carboxylique qui peut être utilisé pour réagir avec les amines primaires via l’activation carbodiimide de l’acide carboxylique. La fluorescéine est le réactif de dérivatisation fluorescent le plus courant pour le marquage des biomolécules. Outre son absorptivité relativement élevée, son excellent rendement quantique de fluorescence et sa bonne solubilité dans l’eau, la fluorescéine possède une excitation maximale qui correspond étroitement à la ligne spectrale de 488 nm du laser argon-ion.
