Coloration fluorescente supérieure
Une analyse de prolifération réussie par dilution de colorant (voir la figure ci-dessous) nécessite un colorant extrêmement lumineux pour distinguer les cellules marquées par fluorescence de l’auto-fluorescence après plusieurs divisions cellulaires. La coloration fluorescente intense procurée par les colorants CellTrace™ CFSE permet la visualisation de huit générations ou plus de cellules proliférant avant que le signal soit submergé par l’auto-fluorescence intrinsèque des cellules. Une coloration uniforme et homogène entraîne très peu de variation de fluorescence entre les cellules d’une population, de sorte que des générations distinctes peuvent être observées sans aucune exigence de logiciel de modélisation complexe.

Rétention du signal à long terme
Contrairement aux colorations qui marquent la membrane lipidique des cellules, le colorant CellTrace™ CFSE traverse facilement la membrane du plasma et se lie de façon covalente à l’intérieur des cellules où le colorant fluorescent, bien retenu et stable, est conçu pour donner un signal constant, même après plusieurs jours dans un environnement de culture cellulaire.

Non toxique
Le colorant CellTrace™ CFSE se lie de façon covalente à toutes les amines libres à la surface et à l’intérieur des cellules et montre peu de cytotoxicité, avec un effet minimal observé sur la capacité de prolifération ou sur la biologie des cellules. Les chercheurs ont utilisé le marquage CFSE SE pour montrer que les cellules hématopoïétiques transplantables prolifèrent in vitro en réponse à une stimulation par un cocktail de facteur de croissance.

Protocole de coloration simple et robuste
Le kit de prolifération cellulaire CellTrace™ contient des flacons de colorant sec jetables pratiques pour permettre de réaliser des expériences à petite échelle sans préparer des quantités excessives de colorant. Une solution mère est préparée en dissolvant le contenu d’un flacon dans du diméthylsulfoxyde (DMSO) anhydre avant l’utilisation. Pour colorer 1 ml de cellules dans un milieu de culture exempt de protéines, il suffit en général d’utiliser 1 µl de cette solution mère. Les cellules doivent être colorées pendant 20 minutes à température ambiante en agitant doucement. Un bref lavage avec du milieu complet pourra alors absorber tout colorant restant dans la solution.
Usage exclusivement réservé à la recherche. Ne pas utiliser pour des procédures de diagnostic.

I'm trying to label my cells in suspension and track them over time by labeling with CFDA SE, but after labeling them they will no longer adhere to a surface (whereas unlabeled cells adhere well). I've tried to label at 10 and 20 µM. What is causing this?

This dye will bind to proteins via primary amines. Too high of a concentration can lead to cell toxicity or unintended modification of cell function. The solution is to reduce the concentration and/or staining time, or to go with a non-protein-binding tracking reagent, such as Qtracker cell labeling reagents.

I want to do a cell migration study for around 4 hours and need to fluorescently label the cells with a dye. What do you recommend?

Calcein, AM and FDA (fluorescein diaceate) are examples of some dyes used for this application. Since these dyes are not incorporated or covalently attached to any cellular components, they may have a short retention time as some cell types may actively efflux the dye out of the cells. The CellTracker and CellTrace dyes include either a mild thiol-reactive chloromethyl group or amine-reactive succinnimidyl ester group to allow for covalent binding to cellular components, providing for better retention. As with any reagent, one should empirically determine retention times for the cell type used.

I labeled my cells with Calcein, AM, but when I imaged the next day, there was no fluorescence from Calcein. Why?

Calcein, AM is a good choice for cell tracking and as a general cytoplasmic stain. However, it doesn't bind to anything and may be actively pumped out of the cells within a couple hours, which is likely what happened. The retention of Calcein within live cells is dependent upon the inherent properties of the cell type and culture conditions.

For long-term imaging, you may wish to consider a reactive cytoplasmic stains such as CFDA, SE or the CellTracker and CellTrace dyes.
