Le FDP est un substrat incolore et non fluorescent pour les phosphatases alcalines. L’hydrolyse séquentielle médiée par les phosphatases alcalines de ses deux substituants de phosphate produit un monophosphate de fluorescéine faiblement fluorescent suivi d’une fluorescéine fortement fluorescente (excitation/émission ∼ 490/514 nm).
