5-CFDA, AM es un sustrato de esterasa que penetra en las células y que puede servir como sonda de viabilidad que mide tanto la actividad enzimática, que es necesaria para activar su fluorescencia, como la integridad célula-membrana, que es necesaria para la retención intracelular de su producto fluorescente. El éster AM electrónicamente neutro de CFDA puede ser cargado en células en concentraciones más bajas que la CFDA. Tras la hidrólisis por esterasas intracelulares, este éster AM también produce carboxifluoresceína. En comparación con la fluoresceína, la carboxifluoresceína contiene cargas negativas adicionales y, por lo tanto, se conserva mejor en las células.
