ITGA4 Antibody (MA1-80529) in ICC/IF
Immunofluorescence analysis of ITGA4 was performed using 70% confluent log phase MOLT-4 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. Clone HP2/1 binds to both intact and the 80kDa fragment of integrin alpha-4. This antibody can be used in basic studies of VLA-4 mediated adhesion and its interaction with the VCAM-1 structure and has been demonstrated to inhibit cell binding to soluble VCAM-1.
This product contains sodium azide; removal by dialysis is recommended prior to use in functional assays.
Mouse anti Human CD49d monoclonal antibody, clone HP2/1 recognizes human CD49d also known as integrin alpha-4 or VLA-4 subunit alpha.
Target Information
The integrin alpha 4 (also known as CD49d and ITGA4) belongs to the integrin alpha chain family of proteins. Integrins are heterodimeric integral membrane proteins composed of an alpha and beta chains. Alpha4 chain associates with either beta1 or beta7 chain. It has been demonstrated that the putative ligand-binding sites of both integrin alpha4beta1 and alpha4beta7 is located on the alpha4 chain. These ligands included Madcam, VCAM, and fibronectin. Madcam is known as the principal ligand for integrin alpha4beta7. Recently it was also demonstrated that HIV-1 envelope can mimic Madcam by binding to and signaling though integrin alpha4beta7, the gut mucosal homing receptor for peripheral T cells.
