Rhodopsin Antibody (MA5-47925) in WB
Western Blot analysis of Rhodopsin was performed using Rhodopsin Recombinant Monoclonal Antibody (Product # MA5-47925). Rhodopsin expressing plasmid transfected (A) and non-transfected (B) HEK293 cells lysate (3 µg protein in RIPA buffer) were resolved on a SDS PAGE gel and blots probed with Rhodopsin Recombinant Monoclonal Antibody at 0.1 µg/mL before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.

Product Specific Information
Specificity: The antibody binds to the C-terminus of rhodopsin on the cytoplasmic side of disk membranes. The epitope has the following amino acid sequence: 'TETSQVAPA'. A limitation of the 1D4 tag, however, is that it has to be placed at the C-terminus of a protein. This is because the Rho1D4 monoclonal antibody requires a free carboxylate group for high affinity binding. Amidation of the carboxyl group lowers its immunoreactivity to the Rho1D4 antibody by over 100 fold.
Target Information
Visual pigments such as rhodopsin and porphyropsin are light-absorbing molecules that mediate vision. Rhodopsin consists of an apoprotein, opsin, covalently linked to 11-cis-retinal. This receptor is coupled to the activation of phospholipase C. Porphyropsin consists of opsin covalently linked to 11-cis 3,4-didehydroretinal.
