Immunocytochemistry analysis of Phospho-N-WASP (Tyr256) in control and pervanadate-treated A431 cells. The cells were labeled with rabbit polyclonal N-WASP/WASP antibody or rabbit Phospho-N-WASP (Tyr256) polyclonal antibody (Product # PA5-143792), then the antibodies were detected using appropriate secondary antibody conjugated to DyLight® 594.

This antibody was cross-adsorbed to phosphotyrosine then affinity purified using phospho-N-WASP (Tyr-256) peptide (without carrier). The antibody detects a 65 kDa* protein corresponding to the molecular mass of phosphorylated N-WASP on SDS-PAGE immunoblots of A431 cells treated with pervanadate. A similar band is observed in pervanadate treated HeLa and endothelial cells. Weak bands are also observed at higher molecular weights after pervanadate treatment. These bands may be due to low cross-reactivity with phosphotyrosine.
Target Information
The Wiskott-Aldrich syndrome (WAS) family of proteins share similar domain structure, and are involved in transduction of signals from receptors on the cell surface to the actin cytoskeleton. The presence of a number of different motifs suggests that they are regulated by a number of different stimuli, and interact with multiple proteins. Recent studies have demonstrated that these proteins, directly or indirectly, associate with the small GTPase, Cdc42, known to regulate formation of actin filaments, and the cytoskeletal organizing complex, Arp2/3. The WASL gene product is a homolog of WAS protein, however, unlike the latter, it is ubiquitously expressed and shows highest expression in neural tissues. It has been shown to bind Cdc42 directly, and induce formation of long actin microspikes.
