PEP-190 is a 16 amino acid synthetic peptide whose sequence corresponds to amino acid residues 1411-1425 from human nNOS. The sequence of PEP-190 is (amino to carboxy terminus) is C N (1411) R L R S E S I A F I E E S K (1425). This sequence is 100% conserved in rat, mouse, frog, and rabbit nNos protiens.
This peptide may be used for neutralization and control experiments with the polyclonal antibody that reacts with this product and rat nNOS, catalog # PA1-033. Using a solution with equal weights per unit volume of peptide and corresponding antibody will yield a solution with a large molar excess of peptide that is able to competitively bind the antibody.
Reconstitute with 0.1 mL of distilled water.
NOS activity requires homodimerization as well as three cosubstrates (L-arginine, NADPH and O2) and five cofactors or prosthetic groups (FAD, FMN, calmodulin, tetrahydrobiopterin and heme). Several distinct NOS isoforms have been described and been shown to represent the products of three distinct genes. These include two constitutive Ca2+/CaM-dependent forms of NOS, including NOS1 (also designated ncNOS) whose activity was first identified in neurons, and NOS3 (also designated ecNOS), first identified in endothelial cells.
