Discover chromatography techniques

Chromatography can separate biomolecules by properties such as charge, hydrophobicity, or affinity. Depending on the molecule, different chromatography techniques may be used during capture, intermediate purification, or polishing.

Affinity chromatography

 

Affinity chromatography is commonly used as the capture step in downstream purification. The target biomolecule, produced in a complex mixture during upstream processing, binds specifically to a ligand on the affinity resin. The target binds tightly to the resin while most process-related impurities either flow through or are washed away. Elution is achieved by applying a buffer that disrupts the binding interaction, releasing the purified target from the column. This technique enables high selectivity and purity early in the purification process, making it one of the most efficient capture methods in bioprocessing.

Hydrophobic Interaction Chromatography (HIC)


 

HIC separates biomolecules based on differences in surface hydrophobicity. In biologics manufacturing, HIC is often used as a complementary polishing step alongside ion exchange chromatography (IEX), helping extend purification power by targeting properties that charge-based methods alone may not fully resolve.

 

Mixed-Mode Chromatography

 

Mixed-mode chromatography utilizes ligands that interact through multiple properties, such as charge and hydrophobicity. This enables the separation of biomolecules that are challenging to resolve with single-mode methods in fewer purification steps.
