SOX1, also known as SRY (sex determining region Y)-box 1, is a transcription factor belonging to the SRY-related HMG box family of transcription factors with a HMG box DNA-binding domain. SOX1 works through multiple independent pathways to promote neuronal cell fate determination and differentiation. This nuclear protein acts as a modulator of LINE retroposons promoter activity and is also involved in the development of the central nervous system, mainly neural and lens development. Transcriptional activator SOX1 binds directly to the HES1 promoter and suppresses HES1 transcription, thus attenuating Notch signaling. It also binds to beta-catenin and suppresses beta-catenin-mediated TCF/LEF signaling, thus potentially attenuating the WNT signaling pathway. Expression of SOX1 is seen mostly in embryonic tissues and moderately in brain.
