          BALB/c mouse Th2-polarized splenocytes were unstimulated (left) or stimulated for 5 hours with the Cell Stimulation Cocktail (plus protein transport inhibitors) (Product # 00-4975-03) (right).           Product Specific Information

          Description
          The 11B11 antibody reacts with mouse interleukin-4 (IL-4), a 14 kD cytokine. The 11B11 antibody is a neutralizing antibody. mouse IL-4, also known as BCDF, BCGF-1 and BSF-1, is secreted by activated Th2 and natural killer T cells, and to a lesser extent by Th1 and mast cells. IL-4 is species-specific and promotes proliferation and differentiation of B cells. IL-4, in conjunction with other signals such as CD40, induces Ig isotype switching of B cells and the upregulation of surface MHC class II and CD23.

          Applications Tested
          This 11B11 antibody has been tested by intracellular staining followed by flow cytometric analysis of mouse Th2-polarized splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol. Please refer to "Staining Intracellular Antigens for Flow Cytometry, Protocol A: Two step protocol for intracellular (cytoplasmic) proteins" located at Flow Protocols. This may be used at less than or equal to 0.25 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

          Blocking Buffers
          When using two or more Super Bright, Brilliant Violet™, Brilliant Ultra Violet™, or other polymer dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75) to minimize any non-specific polymer interactions. Please refer to the datasheet for Super Bright Staining Buffer or Brilliant Stain Buffer for more information.

          Fixation
          • Samples can be stored in IC Fixation Buffer (Product # 00-8222) (100 µL of cell sample + 100 µL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation.
          • Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically.
          • Our internal testing suggests that Brilliant Violet™ 480 (BV480) is not compatible with methanol-based fixation.

          Target Information

          Interleukin-4 (IL-4) is a pleiotropic, immune-modulatory cytokine produced by activated T cells, and serves as a ligand for interleukin 4 receptor. IL-4 has been found to have the broadest range of actions. The interleukin 4 receptor also binds to IL13, which may contribute to many overlapping functions of this cytokine and IL13. STAT6, a signal transducer and activator of transcription, has been shown to play an important role in mediating the immune regulatory signal of IL-4. The IL-4 gene, IL-3, IL-5, IL-13, and CSF2 form a cytokine gene cluster on chromosome 5q. In particular, IL-4 is located in close proximity to IL13 on the chromosome. Further, the IL-4 gene, IL13 and IL5 are found to be coordinated by several long-range regulatory elements over a 120 kilobase range on the chromosome. IL-4 delta 2 is a known splice variant of IL-4.
