          Comparison of immunocytochemistry performance using Parkin Recombinant Rabbit Monoclonal Antibody (21H24L9) (Product # 740019R) and top-cited Parkin antibodies was done. Immunofluorescent analysis of Parkin was performed using 70% confluent SH-SY5Y cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton X-100 for 15 minutes and blocked with 2% BSA for 1 hour at room temperature.           Product Specific Information

          This is an engineered recombinant antibody that was developed to provide superior sensitivity without changing the paratope. Recombinant antibodies are produced using specific genes that code for the desired antibodies. These genes are cloned into an expression vector and expressed in vitro.

          The advantages of recombinant antibodies include better specificity, lot-to-lot consistency and improved signal-to-noise ratio.

          Target Information

          The precise function of Parkin gene is unknown; however, the encoded protein is a component of a multiprotein E3 ubiquitin ligase complex that mediates the targeting of substrate proteins for proteasomal degradation. Mutations in this gene are known to cause Parkinson disease and autosomal recessive juvenile Parkinson disease. Alternative splicing of this gene produces multiple transcript variants encoding distinct isoforms. Additional splice variants of this gene have been described but currently lack transcript support.
