Immunofluorescent analysis of CLCN3 in human neuroblastoma cell line (SK-N-BE). Sample was fixed with 4% formaldehyde (15 min at RT), incubated with CLCN3 monoclonal antibody (Product # MA5-27709) using a dilution of 1:100 (1 hour at RT), and followed by Goat Anti-Mouse 488, Phalloidin Texas Red and DAPI secondary antibody at a dilution of 1:100, 1:1000 (60 min at RT) and 1:5000 (5 min at RT). 1 µg/mL of MA5-27709 was sufficient for detection of Clcn3 in 20 µg of rat brain membrane lysate and assayed by colorimetric immunoblot analysis using goat anti-mouse IgG:HRP as the secondary antibody.|Detects approximately 90kDa. Does not cross-react with Clcn4 or Clcn5 (based on KO validation results).
This antibody was formerly sold as clone S258-5.
Target Information
Mediates the exchange of chloride ions against protons. Functions as antiporter and contributes to the acidification of the endosome and synaptic vesicle lumen, and may thereby affect vesicle trafficking and exocytosis. May play an important role in neuronal cell function through regulation of membrane excitability by protein kinase C. It could help neuronal cells to establish short-term memory.
