          CaV1.2 Antibody (MA5-45387) in ICC/IF

          Immunocytochemistry/Immunofluorescence analysis using human neuroblastoma cells. Fixation involved 4% Formaldehyde for 15 min at RT. Samples were incubated with Cav1.2 monoclonal antibody (Product # MA5-45387) at 1:100 for 60 min at RT, followed by Goat Anti-Mouse ATTO 488 at 1:200 for 60 min at RT. Counterstain used was Phalloidin Texas Red F-Actin stain; DAPI (blue) nuclear stain at 1:1,000, 1:5,000 for 60 min at RT, 5 min at RT. Localization: Cell Membrane, Membrane, Cytoplasm, Nucleoplasm.           Product Specific Information

          1 µg/mL of MA5-45387 was sufficient for detection of Cav1.2 in 10 µg of rat brain lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.|Detects approximately 240kDa (varies with cell background due to glycosylation).

          Target Information

          CACNA1C encodes an alpha-1 subunit of a voltage-dependent calcium channel. Calcium channels mediate the influx of calcium ions into the cell upon membrane polarization. The alpha-1 subunit consists of 24 transmembrane segments and forms the pore through which ions pass into the cell. The calcium channel consists of a complex of alpha-1, alpha-2/delta, beta, and gamma subunits in a 1:1:1:1 ratio. There are multiple isoforms of each of these proteins, either encoded by different genes or the result of alternative splicing of transcripts. The protein encoded by CACNA1C binds to and is inhibited by dihydropyridine. Alternative splicing results in many transcript variants encoding different proteins.
