          CaV3.2 Antibody (MA5-45396) in ICC/IF

          Immunocytochemistry/Immunofluorescence analysis using human neuroblastoma cells. Fixation involved 4% PFA for 15 min. Samples were incubated with Cav3.2 monoclonal antibody (Product # MA5-45396) at 1:50 for overnight at 4°C with slow rocking, followed by AlexaFluor 488 at 1:1,000 for 1 hour at RT. Counterstain used was Phalloidin-iFluor 647 (red) F-Actin stain; Hoechst (blue) nuclear stain at 1:800, 1.6mM for 20 min at RT. (A) Hoechst (blue) nuclear stain.           Product Specific Information

          1 µg/mL of MA5-45396 was sufficient for detection of Cav3.2 in 10 µg of HEK cell lysate expressing Cav3.2 by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.|Detects approximately 260kDa. No cross-reactivity against Cav1.3.

          Target Information

          This gene encodes a T-type member of the alpha-1 subunit family, a protein in the voltage-dependent calcium channel complex. Calcium channels mediate the influx of calcium ions into the cell upon membrane polarization and consist of a complex of alpha-1, alpha-2/delta, beta, and gamma subunits in a 1:1:1:1 ratio. The alpha-1 subunit has 24 transmembrane segments and forms the pore through which ions pass into the cell. There are multiple isoforms of each of the proteins in the complex, either encoded by different genes or the result of alternative splicing of transcripts. Alternate transcriptional splice variants, encoding different isoforms, have been characterized for the gene described here. Studies suggest certain mutations in this gene lead to childhood absence epilepsy (CAE).
