          LRRK2 Antibody (MA5-45643) in ICC/IF

          Immunocytochemistry/Immunofluorescence analysis using human neuroblastoma cells. Fixation involved 4% Formaldehyde for 15 min at RT. Samples were incubated with LRRK2/Dardarin monoclonal antibody (Product # MA5-45643) at 1:100 for 60 min at RT, followed by Goat Anti-Mouse ATTO 488 at 1:100 for 60 min at RT. Counterstain used was Phalloidin Texas Red F-Actin stain; DAPI (blue) nuclear stain at 1:1,000, 1:5,000 for 60 min RT, 5 min RT. Localization: Cytoplasm. Magnification: 60X.           Product Specific Information

          81% identical in mouse, 80% identical in rat. <30% identity with LRRK1.

          10 µg/mL of MA5-45643 was sufficient for detection of LRRK2/Dardarin in 20 µg of rat brain lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.|Detects >200kDa.

          This antibody was formerly sold as clone S231B-34.

          Target Information

          LRRK2 is a multidomain protein that includes a kinase domain, a GTPase domain, and a leucine-rich repeat (LRR) domain. Mutations in LRRK2 have been associated with Parkinson's disease. LRRK2 is a member of the leucine-rich repeat kinase family and encodes a protein with an ankryin repeat region, a leucine-rich repeat domain, a kinase domain, a DFG-like motif, a RAS domain, a GTPase domain, a MLK-like domain, and a WD40 domain. The protein is present largely in the cytoplasm but also associates with the mitochondrial outer membrane.
