Video: How to perform neural induction with Gibco PSC Neural Induction Medium

PSC Neural Induction Medium workflow

schematic for using Gibco PSC Neural Induction Medium for neural induction in 7 days

A simple protocol is used to initiate PSC induction to neural stem cells in only 7 days.

Neural induction workflow and timeline:

Day minus 1
  • Prepare complete Neural Induction Medium
  • Split and plate PSCs

Day 0

  • Exchange medium for complete PSC Neural Induction Medium

Days 1–6

  • Check cells, change medium every other day until cells reach 100% confluency

Day 7

  • Harvest NSCs, proceed with disassociation

Cells are now ready for expansion and terminal neuronal differentiation.

Accelerate your neural inductions with Gibco Neural Induction Medium

Figure 1. Gibco PSC Neural Induction Medium does not require the intermediary step of embryoid body (EB) formation, helping you save time and labor when generating NSCs.

"In this study, we report a highly efficient serum-free pluripotent stem cell neural induction medium that can induce hPSCs into primitive NSCs (pNSCs) in 7 days, obviating the need for time-consuming, laborious embryoid body generation or rosette picking."

Achieve high-efficiency neural induction with PSC Neural Induction Medium

fluorescence microscopy of Oct4, Nestin, Sox1, and Sox2; graph showing % of positive cells for each marker

Figure 2. NSCs grown in PSC Neural Induction Medium express neural markers and have 80–90% neural induction efficiency. NSCs derived from hPSCs were fixed and flabeled with antibodies against pluripotent marker Oct4, neural marker Nestin (red), Sox1 and Sox2 (green). Cell nuclei were stained with DAPI (blue). Quantitative analysis of the percentage of positive cells to Nestin, Sox1, and Sox2 shows 80–90% neural induction efficiency.

Figure 3.NSCs grown in PSC Neural Induction Medium have differentiation potential. NSCs generated using Gibco PSC Neural Induction Medium demonstrate capacity to be differentiated into neurons (expressing beta III tubulin), astrocytes (GFAP), or oligodendrocytes (GalC).
