Dual fluorescence and brightfield cell counting
 

The lnvitrogen Countess 3 FL Automated Cell Counter has fluorescence capabilities with two easily interchangeable fluorescent light cubes selected from 13 spectrum-spanning options. The Countess 3 FL Automated Cell Counter can count cells and measure viability in fluorescent mode as well as brightfield, while also reporting fluorescent protein expression, apoptosis, transfection efficiency, and cell viability using fluorescent reagents in as few as 30 seconds. Fluorescence cell counting also allows the use of fluorescence-based cellular assays that offer more specificity with complex samples by using fluorescent cellular stains or fluorescently tagged cells. With the utilization of artificial intelligence during development, our team has applied deep-learning neural network algorithms to generate highly accurate cell counts, even for the most challenging cell sample types.

Fluorescence counting applications

Go beyond cell counting with the integrated fluorescence capabilities of the Countess 3 FL Automated Cell Counter. Easily assess cell viability, apoptosis, transfection efficiency, and fluorescent protein expression—while having the flexibility to analyze virtually any cellular parameter that is autofluorescent or dye-labeled. Autofocus and autolighting

Autofocus can easily detect and focus on cells enabling accurate counts. Autolighting in both the brightfield and fluorescent channels enable optimal images. Users can effortlessly adjust the focus and lighting for difficult samples and save the autofocus start point.

Fluorescence intensity excitation lock

When running many similar samples back-to-back, locking the fluorescence intensity reduces workflow time and increases consistency between subsequent counts.

Detect more with fluorescence-based counting

Increased sensitivity enables identification of lower-intensity signals, with flexible gating to refine results. The fluorescence-based counting mode expands flexibility by identifying and counting objects directly from fluorescent images, rather than relying on brightfield analysis. This approach can detect lower-intensity fluorescent objects, increasing sensitivity and enabling more comprehensive analysis—while still allowing users to gate out unwanted signals.

 

Because brightfield and fluorescence-based counts analyze samples differently, results such as object counts and size measurements may vary, giving users complementary insights depending on their experimental needs.


Interchangeable LED light cubes

The Countess 3 FL Automated Cell Counter has two optional fluorescence channels, which will detect fluorescent colors determined by the insertion of individual light cubes. The Countess 3 FL has the greatest number of fluorescent color options available of any benchtop automated cell counter with 13 interchangeable EVOS LED light cubes to choose from, enabling detection of cells far beyond GFP and RFP. Each cube contains an LED, illuminating optics, and filters. Light cubes are user-interchangeable and automatically detected by the Countess 3 FL Automated Cell Counter for plug-and-play ease. Additionally, the light cubes are interchangeable with the entire line of EVOS imaging systems, minimizing the cost of both systems.
