IL-4 Antibody (48-7049-42) in Flow
Normal human peripheral blood cells were stimulated for 5 hours with the Cell Stimulation Cocktail (plus protein transport inhibitors) (Product # 00-4975-93) and then stained intracellularly, using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol, with CD3 Monoclonal Antibody, APC (Product # 17-0038-42) and Mouse IgG1 kappa Isotype Control, eFluor 450 (Product # 48-4714-82) (left) or IL-4 Monoclonal Antibody, eFluor 450 (right). Cells in the lymphocyte gate were used for analysis.

Description: The 8D4-8 antibody reacts with human interleukin-4 (IL-4), a 15-19 kDa cytokine secreted by Th2 cells.
Applications Reported: This 8D4-8 antibody has been reported for use in intracellular staining followed by flow cytometric analysis.
Applications Tested: This 8D4-8 antibody has been pre-diluted and tested by flow cytometric analysis of stimulated normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol. Please refer to "Staining Intracellular Antigens for Flow Cytometry, Protocol A: Two step protocol for intracellular (cytoplasmic) proteins" located at www.thermofisher.com/flowprotocols . This may be used at 5 µL (0.25 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.
eFluor 450 is an alternative to Pacific Blue. eFluor 450 emits at 446 nm and is excited with the violet laser line (405 nm). Please make sure that your instrument is capable of detecting this fluorochrome.

Target Information
Interleukin-4 (IL-4) is a pleiotropic, immune-modulatory cytokine produced by activated T cells, and serves as a ligand for interleukin 4 receptor. IL-4 has been found to have the broadest range of actions. The interleukin 4 receptor also binds to IL13, which may contribute to many overlapping functions of this cytokine and IL13. STAT6, a signal transducer and activator of transcription, has been shown to play an important role in mediating the immune regulatory signal of IL-4. The IL-4 gene, IL-3, IL-5, IL-13, and CSF2 form a cytokine gene cluster on chromosome 5q. In particular, IL-4 is located in close proximity to IL13 on the chromosome. Further, the IL-4 gene, IL13 and IL5 are found to be coordinated by several long-range regulatory elements over a 120 kilobase range on the chromosome. IL-4 delta 2 is a known splice variant of IL-4.
