Demonstrate sequential development of germ tubes and chlamydospores by Candida albicans with Thermo Scientific™ Remel™ RIOT Agar (Rice Infusion, Oxgall, Polysorbate 80). In 1968, Fischer et al. modified rice infusion agar of Taschdian by adding 2% oxgall to the medium1. In 1975, Beheshti et al. formulated a medium containing rice infusion, oxgall and polysorbate 80 for sequential development of germ tubes and chlamydospores, based on the formulas of Taschdjian and Fischer2. This medium was called RIOT Agar.

RIOT Agar (Rice Infusion, Oxgall, Polysorbate 80) is a solid medium that is used to demonstrate sequential development of germ tubes and chlamydospores by Candida albicans.

This medium contains oxgall and polysorbate 80 that stimulates the formation of germ tubes and chlamydospores.

  • Ready-to-use – Convenience of prepared media.
  • Easy-to-interpret
    • Germ tube production – A short hyphal (filamentous) extension from a yeast cell, with no constriction at the point of origin indicates a positive test. No short hyphal (filamentous) extension indicates a negative test.
    • Chlamydospore production – Clusters of round blastoconidia at the septa and large, thick-walled terminal chlamydospores indicate a positive test. No clusters of round blastoconidia or chlamydospores indicate a negative test.

RIOT Agar (Rice Infusion, Oxgall, Polysorbate 80) contains cream of rice that provides essential nutrients which support growth of Candida spp. Oxgall and polysorbate 80 stimulate the formation of germ tubes and chlamydospores. Agar is the solidifying agent.
