Allgemeine Beschreibung
• Fluconazol, 98 %, ist eine Triazol-Verbindung, die hauptsächlich gegen Candida-, Coccidioides- und Cryptococcus-Arten eine antimykotische Wirkung zeigt
•Es konnte gezeigt werden, dass es die Sterol-C-14α-Demethylierung von Cytochrom P-450 in Pilzen hemmt, was zur Ansammlung von 14α-Methylsterolen in Pilzen, dem Verlust normaler Pilzsterole und der damit verbundenen fungistatischen Aktivität führt
•Hemmt zudem Cytochrom P450-Familie 2, Unterfamilie c, Mitglied 9 (CYP2C9)
•Die Demethylierung von Säugetierzellen ist deutlich weniger anfällig für die Fluconazol-Hemmung
Anwendungen
• Fluconazol weist eine fungistatische Aktivität gegen viele Pilzstämme auf, darunter Candida albicans, Candida glabrata, Candida parapsilosis, Candida tropicalis und Cryptococcus neoformans.
•Fluconazol beeinträchtigt die Ergosterol-Synthese in Pilzen und reguliert das Metallothionein-Gen nach unten
•In biologischen Experimenten wurde diese Verbindung zur Beurteilung der minimalen Inhibitorkonzentration in (MIC-)Assays von Pilzproben verwendet
• Wurde auch als Positivkontrolle bei antimykotischen Assays von Pflanzen verwendet
