iBlot 2 Transfer Stacks are used to transfer proteins using the iBlot 2 Gel Transfer Device. They are consumable stacks with an integrated pre-activated PVDF transfer membrane for dry blotting of proteins. Each iBlot 2 Transfer Stack contains a copper-coated electrode and appropriate cathode and anode buffers in the gel matrix to allow fast, reliable transfer of proteins. This mini-size stack (8 x 8 cm) can be used with one mini-size gel.

PVDF membrane
The PVDF membrane (0.2-μm pore size, low fluorescence) in this stack has higher binding capacity than nitrocellulose. It is pre-activated and ready for use without any pretreatment with alcohols. The membrane is compatible with commonly used detection methods such as staining, immunodetection, fluorescence, and radiolabeling. The proteins bind to the membrane due to hydrophobic interactions. The protein-binding capacity is 240 μg/cm2.

PVDF-Membran
Die PVDF-Membran (0,2 µm-Porengröße, geringe Fluoreszenz) in diesem Stapel hat ein höheres Bindungsvermögen als Nitrozellulose. Sie ist voraktiviert und ohne Vorbehandlung mit Alkoholen gebrauchsfertig Die Membran ist kompatibel mit gängigen Nachweismethoden, z. B. Färbung, Strukturanalyse, Fluoreszenz und Radiomarkierung. Die Proteine binden aufgrund von hydrophoben Wechselwirkungen an die Membran. Die Protein-Bindungskapazität beträgt 240 μg/cm².

If a green color appears on the membrane after the transfer using the iBlot 2 Dry Blotting System, will it affect the results?

The green discoloration is copper deposits from the transfer stack, and it does not affect the results. To minimize this effect, shake excess water off the filter paper and buffer from the gel before placing each on the stack. The current formulation of stacks minimizes the green discoloration.

Can I cut the bottom stack of my iBlot 2 Transfer Stack to fit my separating gel?

No. Do not trim the iBlot 2 Transfer Stacks to fit your gel size.

The PVDF membrane is pre-activated and ready for use without any pre-treatment with alcohol.

Can I use the stack and/or absorbent pad more than once when using the iBlot 2 Dry Blotting System?

No. The transfer stacks have a finite amount of ions to drive the transfer and are depleted after a single use.

Can I perform multiple transfers, with a new transfer starting immediately after the previous one when using the iBlot 2 Dry Blotting System?

Yes. The device is designed to do so with no impact on performance.
