Poly-D-Lysine is a chemically synthesized extracellular matrix used to facilitate cell adhesion to tissue culture-treated plastic and glass surfaces. Poly-D-Lysine contains polymers with a molecular weight of 50,000–150,000 Daltons, which makes this product ideal for neuronal culture applications. This ready-to-use product is provided as a liquid at a concentration of 0.1 mg/mL. When used on Nunc cell culture plastics with B-27 supplements or the B-27 Plus Neuronal Culture System, Poly-D-Lysine maintains cell viability and normal growth characteristics in multiple neuronal cell types.

I coated my plates with Poly-D-Lysine and I'm noticing that the cells are dying within a few days of plating the cells. How can I ensure cell survival?

It is possible that the plates were not adequately rinsed with water prior to plating the cells. Residual Poly-D-Lysine is toxic and may be the cause of cell death. It is recommended to rinse the plates with water three times before exposing the cells to the matrix.
