          BALB/c mouse splenocytes were stimulated for 72 hours with CD3e Monoclonal Antibody, Functional Grade (Product # 16-0031-85), CD28 Monoclonal Antibody, Functional Grade (Product # 16-0281-85), Mouse IL-2 Recombinant Protein, and Mouse IL-4 Recombinant Protein.           Product Specific Information

          Description
          The eBio13A antibody reacts with mouse IL-13. IL-13 is a cytokine produced mainly by Th2 cells, but also by antigen-primed CD8 T cells. IL-13 has a strong involvement in allergic inflammation and parasitic clearing and in cancer models has been shown to have either inhibitory or stimulatory activity depending on the tumor. In humans, IL-13 is found to play a role in isotype switching in B cells. IL-13 is implicating in down modulating macrophage activity, through the reduction of pro-inflammatory cytokines (IL-1, IL-6, IL-8, IL-10, IL-12).

          Applications Tested
          This eBio13A antibody has been tested by intracellular staining followed by flow cytometric analysis of stimulated mouse splenocytes. This may be used at less than or equal to 1.0 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

          Blocking Buffers
          When using two or more Super Bright, Brilliant Violet™, Brilliant Ultra Violet™, or other polymer dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75) to minimize any non-specific polymer interactions. Please refer to the datasheet for Super Bright Staining Buffer or Brilliant Stain Buffer for more information.

          Light sensitivity
          This tandem dye is sensitive to photo-induced oxidation. Please protect this vial and stained samples from light.

          Fixation
          • Samples can be stored in IC Fixation Buffer (Product # 00-8222) (100 µL of cell sample + 100 µL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation.
          • Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically.
          • Our internal testing suggests that Brilliant Ultra Violet™ 805 (BUV805) is compatible with short-term methanol-based fixation, but should not be stored in buffers containing methanol for longer than one hour.

          Target Information

          IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells, and is involved in regulating inflammatory and immune responses. IL-13 down-regulates macrophage activity, thereby inhibiting the production of pro-inflammatory cytokines and chemokines, and is involved in allergen-induced asthma through mechanisms independent of IgE and eosinophils. IL-13 also regulates several stages of B cell maturation and differentiation, up-regulates CD23 and MHC class II expression, and promotes IgE isotype switching of B cells. Structurally, IL-13 closely resembles IL-3, IL-4, IL-5 and GM-CSF. The gene encoding IL-13, IL3, IL5, IL4, and CSF2 form a cytokine gene cluster on chromosome 5q, with the IL-13 gene particularly close to IL4.
