BALB/c mouse splenocytes were stimulated for 72 hours with CD3e Monoclonal Antibody, Functional Grade (Product # 16-0031-85), CD28 Monoclonal Antibody, Functional Grade (Product # 16-0281-85), Mouse IL-2 Recombinant Protein, and Mouse IL-4 Recombinant Protein. Description
The eBio13A antibody reacts with mouse IL-13. IL-13 is a cytokine produced mainly by Th2 cells, but also by antigen-primed CD8 T cells. IL-13 has a strong involvement in allergic inflammation and parasitic clearing and in cancer models has been shown to have either inhibitory or stimulatory activity depending on the tumor. In humans, IL-13 is found to play a role in isotype switching in B cells. IL-13 is implicating in down modulating macrophage activity, through the reduction of pro-inflammatory cytokines (IL-1, IL-6, IL-8, IL-10, IL-12).
Applications Tested
This eBio13A antibody has been tested by intracellular staining followed by flow cytometric analysis of stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol. Please refer to "Staining Intracellular Antigens for Flow Cytometry, Protocol A: Two step protocol for intracellular (cytoplasmic) proteins" located at Flow Protocols . This may be used at less than or equal to 1.0 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Blocking Buffers
When using two or more Super Bright, Brilliant Violet™, Brilliant Ultra Violet™, or other polymer dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75) to minimize any non-specific polymer interactions. Please refer to the datasheet for Super Bright Staining Buffer or Brilliant Stain Buffer for more information.
Light sensitivity
This tandem dye is sensitive to photo-induced oxidation. Please protect this vial and stained samples from light.
Fixation
• Samples can be stored in IC Fixation Buffer (Product # 00-8222) (100 µL of cell sample + 100 µL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation.
• Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically.
• Our internal testing suggests that Brilliant Ultra Violet™ 737 (BUV737) is compatible with short-term methanol-based fixation, but should not be stored in buffers containing methanol for longer than one hour.

Target Information
IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells, and is involved in regulating inflammatory and immune responses. IL-13 down-regulates macrophage activity, thereby inhibiting the production of pro-inflammatory cytokines and chemokines, and is involved in allergen-induced asthma through mechanisms independent of IgE and eosinophils. IL-13 also regulates several stages of B cell maturation and differentiation, up-regulates CD23 and MHC class II expression, and promotes IgE isotype switching of B cells. Structurally, IL-13 closely resembles IL-3, IL-4, IL-5 and GM-CSF. The gene encoding IL-13, IL3, IL5, IL4, and CSF2 form a cytokine gene cluster on chromosome 5q, with the IL-13 gene particularly close to IL4.
