IP-Lysepuffer ist ein Säugetier-Ganzzelllyse-Reagenz, das auf einer modifizierten RIPA-Pufferformulierung ohne SDS basiert. Dieser Lysepuffer zeichnet sich durch moderate Stärke aus. Er bewirkt eine effektive Solubilisierung der Zellproteine, ohne genomische DNA freizusetzen oder Proteinkomplexe aufzubrechen wie normaler RIPA-Puffer. Pierce IP Lysis Buffer wurde speziell für Pulldown-Assays und Immunpräzipitationsassays entwickelt.

Pierce IP Lysis Buffer ist für das Lysieren kultivierter Säugetierzellen aus sowohl plattierten Zellen als auch Zellen, die aus Suspensionskulturen pelletiert werden, geeignet. Dieser Lysepuffer wurde für Pulldown- und Immunpräzipitationsassays optimiert und ist auch mit vielen anderen Anwendungen kompatibel, darunter Thermo Scientific Pierce BCA- und 660-nm-Proteinassays, Proteinaufreinigung und Immunassays (z. B. ELISA, Western Blot).

Pierce IP Lysis Buffer besteht aus 25 mM Tris-HCl pH 7,4, 150 mM NaCl, 1 mM EDTA, 1 % NP-40 und 5 % Glycerin. Der Puffer enthält keine Protease- oder Phosphatase-Inhibitoren; auf Wunsch können jedoch Inhibitoren wie Thermo Scientific Halt Protease Inhibitor Cocktail oder Phosphatase Inhibitor Cocktail unmittelbar vor der Verwendung hinzugefügt werden, um eine Proteolyse zu verhindern und die Phosphorylierung von Proteinen aufrechtzuerhalten.

The most commonly used buffer is RIPA Buffer with SDS. We offer RIPA Buffer (Cat. Nos. 89900 and 89901). We also offer the Pierce IP Lysis buffer (Cat. Nos. 87787 and 87788) as well as M-PER (Cat. Nos. 78501, 78503, and 78505).

Pierce IP Lysis Buffer ships at ambient temperature and can be stored at room temperature. However, we recommend 4 degrees C for long term storage in order to prevent possible bacterial growth.

Which protein assays are compatible with the Thermo Scientific IP Lysis Buffer?

The Thermo Scientific IP Lysis Buffer is compatible with Thermo Scientific BCA Protein Assay (Cat. No. 23225) and Thermo Scientific 660 nm Protein Assay (Cat. No. 22660). It is not compatible with Thermo Scientific Coomassie Plus (Bradford) Assay (Cat. No. 23236) unless diluted 1:1 in TBS, PBS, or water.
