          Phospho-PTP1B (Ser50) Antibody (PA5-143754) in ICC/IF

Immunocytochemistry analysis of Phospho-PTP1B (Ser50) in aldehyde-fixed and NP-40 permeabilized human NCI-H1915 lung carcinoma cells. The cells were labeled with rabbit polyclonal anti-Phospho-PTP1B (Ser50) polyclonal antibody (Product # PA5-143754). The antibody was detected using appropriate secondary antibody conjugated to DyLight® 594. {{ $ctrl.currentElement.advancedVerification.fullName }} validation info. View more

PTP1B (Ser-50) synthetic peptide (coupled to carrier protein) corresponds to amino acids surrounding Ser-50 in human PTP1B. This sequence has significant homology to the conserved site in rat and mouse PTP1B, and has homology to conserved sites in TC-PTP and MPTP, but not other protein tyrosine phosphatase family members.

          Product Specific Information

The antibody detects a 50 kDa* doublet corresponding to PTP1B in western blots of calyculin A treated human A431 and Jurkat cells. This reactivity is observed as a weak singlet in control cells and is not observed after lambda phosphatase treatment.

          Target Information

PTP1B (PTPN1) is a tyrosine-protein phosphatase which acts as a regulator of endoplasmic reticulum unfolded protein response. It mediates dephosphorylation of EIF2AK3/PERK and in turn inactivates the protein kinase activity of EIF2AK3/PERK. PTP1B also plays a role in CKII- and p60c-src-induced signal transduction cascades. It regulates the EFNA5-EPHA3 signaling pathway which modulates cell reorganization and cell-cell repulsion. PTP1B also regulates the hepatocyte growth factor receptor signaling pathway through dephosphorylation of MET.
