Thermo Scientific Pierce Sulfo-SMCC, No-Weigh Format is a water-soluble, amine-to-sulfhydryl crosslinker that contains NHS-ester and maleimide reactive groups at opposite ends of a medium-length cyclohexane spacer arm (8.3 angstroms).

Features of Sulfo-SMCC, No-Weigh Format:

Reactive groups: sulfo-NHS ester and maleimide
Reactive towards: amino and sulfhydryl groups
• Water-soluble (compare to SMCC), so crosslinking can be done in physiologic solutions
• High purity, crystalline reagent can be used to create high-purity maleimide-activated derivatives
• Cyclohexane bridge confers added stability to the maleimide group making Sulfo-SMCC the ideal crosslinking agent for maleimide activation of proteins. Maleimide groups are stable for 64 hours in 0.1 M sodium phosphate buffer, pH 7 at 4°C.

Sulfosuccinimidyl-4-(N-maleimidomethyl)cyclohexane-1-carboxylate (Sulfo-SMCC) is a non-cleavable and membrane impermeable crosslinker. It contains an amine-reactive N-hydroxysuccinimide (NHS ester) and a sulfhydryl-reactive maleimide group. NHS esters react with primary amines at pH 7-9 to form stable amide bonds. Maleimides react with sulfhydryl groups at pH 6.5-7.5 to form stable thioether bonds. The maleimide groups of Sulfo-SMCC and SMCC are unusually stable up to pH 7.5 because of the cyclohexane bridge in the spacer arm. Because it contains the hydrophilic sulfonyl moiety, Sulfo-SMCC is soluble up to ∼10 mM in water and many commonly used buffers, thus avoiding the use of organic solvents which may perturb protein structure.

• Enzyme labeling of antibodies - both enzyme and antibody specificity can be preserved
• Create specific bioconjugates via one- or two-step crosslinking reactions
• Create sulfhydryl-reactive, maleimide-activated carrier proteins for coupling haptens

How does a product with "No-Weigh" format reduce my concern over reagent stability?

The No-Weigh product format, where small amounts of reagent are pre-weighed and packaged separately, prevents the contamination and loss of reagent reactivity over time associated with repetitive opening and closing of a single vial. This format enables use of a fresh vial of reagent each time and eliminates the hassle of weighing out small amounts of reagent. We are using EDC to crosslink a protein onto carboxyl-modified beads. We would like to add in a linker molecule to extend the radius of beads to increase the surface area so more proteins can be immobilized. Is this possible?

To provide a linker between the beads and the target protein, you would need a molecule with a primary amine (to conjugate to the carboxyl group on the beads with EDC) on one end and another reactive group on the other (to conjugate to the protein). For instance, sulfo-SMCC (Cat. No. A39268) could be used in conjunction with PDPH (Cat. No. 22301):
The method is outlined below:
1. Conjugate the target protein to the NHS ester (amine-reactive) group of Sulfo-SMCC. This will produce a maleimide-activated target protein via its primary amines (lysines and N-terminus).
2. Remove the excess sulfo-SMCC by desalting or dialysis.
3. React the maleimide-activated target protein to PDPH via sulfhydryls. (Lysines and N-terminus now are connected to hydrazide (amine) group with an extended linker)
4. Remove excess PDPH by desalting or dialysis.
5. React the hydrazide-activated target protein with carboxyl beads in the presence of EDC.
6. Use appropriate buffer to rinse off excess target protein from the carboxyl beads.
