About MAX Efficiency DH5α Competent Cells
MAX Efficiency DH5α Competent Cells have been prepared by a proprietary modification of the procedure of Hanahan (1983). These cells are suitable for the construction of gene banks or for the generation of cDNA libraries using plasmid-derived vectors. The Φ80lacZΔM15 marker provides α-complementation of the β-galactosidase gene from pUC or similar vectors, and can therefore be used for blue/white screening of colonies on bacterial plates containing Bluo-gal or X-gal. DH5α cells are capable of being transformed efficiently with large plasmids, and can also serve as a host for the M13mp cloning vectors if a lawn of DH5α-FT, DH5αF′, DH5αF′IQ, JM101, or JM107 is provided to allow plaque formation.

Library Efficiency DH5α Competent Cells are a versatile strain of chemically competent cells that demonstrate transformation efficiencies of >1 x 108 cfu/µg plasmid DNA. Library Efficiency DH5α Competent Cells are ideal for difficult cloning constructions or any application that uses limiting amounts of DNA. Features of these cells:

• Supports blue/white screening by α-complementation on plates containing X-Gal or Bluo-Gal
• Maintains plasmid DNA with minimum recombination events
• Delivers high-yield plasmid preparations for downstream applications

Ideal for difficult cloning projects
Library Efficiency DH5α Competent Cells are recommended for routine cloning of genes into large plasmids, and are suitable for difficult clone constructions, such as blunt-end ligations. Library Efficiency DH5α Competent Cells have a mid-range transformation efficiency (>1 x 108 cfu/µg control DNA per 100 µL reaction). The strain also has the Φ80lacZΔM15 genotype, allowing blue/white screening on plates containing either X-Gal or Bluo-Gal. Finally, the recA1 mutation helps reduce the rate of recombination while propagating plasmid DNA, and the endA1 mutation makes DH5α cells an excellent choice for amplifying plasmid DNA for subsequent extraction and purification.

One Shot™ TOP10F´ Chemically Competent E. coli (Invitrogen™)

One Shot® TOP10F´ Chemically Competent E. coli are identical to TOP10 cells, with the addition of an F´ episome. TOP10 competent cells are provided at a transformation efficiency of 1 x 109 cfu/µg supercoiled DNA and are ideal for high-efficiency cloning and plasmid propagation.

Using One Shot® TOP10F´ Chemically Competent E. coli
The F´ episome carries the tetracycline resistance gene and allows isolation of single-stranded DNA from vectors that have an f1 origin of replication. In addition, the F´ episome carries the lacIq repressor for inducible expression from trc, tac, and lac promoters using IPTG. TOP10F´ cells require IPTG induction for blue/white color screening.

DH10B cells are available in both electrocompetent and chemically competent formats. Our ElectroMax DH10B T1R Cells and MegaX DH10B T1R Electrocomp Cells offer the benefit of T1 and T5 phage resistance.

Competent Cell Sampler (Invitrogen™)

The Competent Cell Sampler kit contains a pair of four different strains of chemically competent E. coli cells in convenient One Shot formats. Each single-use vial of cells is ready to use, with no extra pipetting steps or freeze-thaw cycles which reduce transformation efficiency. Use this kit to evaluate which strains best suit the different types of DNA used in your lab.

One Shot Mach1 T1-phage resistant chemically competent E. coli cells have a faster doubling time than typical E. coli cells. As a result, Mach1 colonies are clearly visible within eight hours of plating your transformation on ampicillin selection plates. From an overnight colony, minipreps can be performed after four hours of growth.

Mach1 competent E. coli cells come in One Shot format (Cat. No. C862003) and our MultiShot StripWell and FlexPlate formats for medium and high-throughput applications (C869601 and C8681201).

One Shot OmniMAX2 T1-phage-resistant chemically competent E. coli cells are perfect for use in all cloning applications, including Gateway technology. They offer the highest transformation efficiency in a chemically competent One Shot format with 5 x 109 transformants with 1 µg of control DNA. Coupling the high transformation efficiency with the elimination of the mcrA, mrr, mcrBC, and hsdRMS restriction systems allows for the construction of more representative genomic libraries in a convenient chemically competent format.

OmniMAX 2 cells are offered in our One Shot format (C854003) and in our MultiShot FlexPlate format for medium to high throughput applications (C8581201).

OneShot Stbl3 chemically competent cells reduce the frequency of unwanted homologous recombination and are perfect for cloning unstable DNA such as lentiviral DNA containing long terminal repeats (LTRs). The DNA yield from Stbl3 cells is often >10-fold higher than standard cloning strains.

Stbl3 chemically competent cells are available in the One Shot format (C737303) and in the MultiShot StripWell and FlexPlate formats (C739601 and C7381201) for medium to high throughput applications.

One Shot TOP10 cells are perfect for a broad range of cloning techniques. They are often included with many of our cloning and expression kits like TOPO and Gateway cloning kits and complement our GeneArt High-Q Strings and Strings DNA fragments and libraries. One Shot TOP10 competent E. coli come with a transformation efficiency of 1 x 109 cfu/µg supercoiled DNA and are ideal for high-efficiency cloning and plasmid propagation. They allow stable replication of high-copy number plasmids.

TOP10 chemically competent cells are available in the One Shot format (C404003), in the MultiShot StripWell and FlexPlate formats (C409601 and C4081201) for medium to high throughput applications.

One Shot™ TOP10 Electrocomp™ E. coli (Invitrogen™)

One Shot® TOP10 Electrocomp™ E. coli are provided at a transformation efficiency of 1 x 1010 cfu/ µg supercoiled DNA and are ideal for high-efficiency cloning and plasmid propagation. They allow stable replication of high copy number plasmids. The genotype of TOP10 cells is similar to that of the DH10B™ strain, and offers the following features:

hsdR for efficient transformation of unmethylated DNA from PCR amplifications
mcrA for efficient transformation of methylated DNA from genomic preparations
lacZΔM15 for blue/white color screening of recombinant clones
endA1 for cleaner preparations of DNA—get better results in downstream applications due to the elimination of nonspecific digestion by Endonuclease I
recA1 for reduced occurrence of nonspecific recombination in cloned DNA
