Accurate cell viability data in less than 30 seconds

The Countess 3 FL Automated Cell Counter has fluorescence capabilities with two fluorescence channels that can easily be changed with 15 light cubes to choose from, offering class-leading flexibility. With the option for fluorescence capabilities, the Invitrogen Countess 3 FL Automated Cell Counter can count cells in brightfield, while also reporting and measuring fluorescent protein expression, apoptosis, and cell viability using fluorescent reagents in as little as 30 seconds. Fluorescence cell counting should be considered when using fluorescence cellular assays that will provide more specificity to complex samples and/or sensitivity for low-abundance or hard-to-detect cells.

The Countess 3 FL Automated Cell Counter is a highly accurate brightfield and dual fluorescence cell counter, with interchangeable light cubes.


Fluorescence capabilities—select any two fluorescence colors from 15 light cube options

The Countess 3 FL instrument has two optional fluorescence channels. The fluorescent colors collected are determined by the insertion of individual light cubes. With 15 light cubes to choose from, the Countess 3 FL instrument is not limited to detection of GFP- and RFP-expressing cells. Interested in measuring apoptosis and viability of cells in a population? No problem. Use an apoptosis indicator that measures caspase activation, such as Invitrogen CellEvent Caspase-3/7 Green Detection Reagent along with the EVOS GFP Light Cube. Additionally, use a viability dye such as Invitrogen SYTOX Red Dead Cell Stain with the EVOS Cy5 Light Cube to measure dead cells that have progressed past apoptosis. This staining combination along with the two EVOS light cubes offers a fast and simple method to obtain apoptosis and viability data.

Cell viability assessment

Assessing cell viability is a key step in daily cell culture and is required for accurate and efficient downstream processing. With the Countess 3 FL instrument, there are fluorescence options that offer a sensitive and specific determination of viability. These can be done quickly and easily to check viability prior to downstream sample analysis. By using Invitrogen ReadyCount Green/Red Viability Stain which contains acridine orange and propidium iodide (AO/PI) stains in combination, the fluorescence staining provides great sensitivity and greater specificity of the live/dead status amid a mixed cell population, and it does not alter the morphology of the cell (Figure 1).

Measure transfection efficiency

The ability to assess the number of cells successfully transfected or transduced in a cell population is a basic and critical evaluation parameter. Commonly, transfection or transduction of cells of interest results in the expression of a fluorescent protein reporter, such as GFP. The Countess 3 FL Automated Cell Counter, loaded with your choice of light cubes, offers a quick and simple method to easily obtain transfection efficiency data (Figure 2).

The result, image, and data files can be easily saved to a USB drive or by Wi-Fi enabled connectivity to the Cloud using Thermo Fisher Connect. There are five different files that can be saved. Autosave enables automatic saving of data, so there is no need to select Save after each count.

  • The “Result” and “Images” files can be saved as JPEG, TIFF, or PNG files.
  • The “Report” file is a PDF file and includes the result and image files.
  • Data files are in CSV format, which:
    • can be easily imported into your current analysis software or viewed in Excel software.
    • has the option to save All, Last 30 Days, Last 7 Days, Today, or Current.
  • The “Cell Data” file is an FCS file for advanced individual cell data plotting and analysis.
