TaqMan Fast Advanced Master Mix supports optimal efficiency in duplex reactions
Achieving the optimal 90–110% efficiency for both assays in a duplex reaction helps ensure accurate quantification of both targets with minimal bias. A FAM-labeled assay (ACTB) and a VIC-labeled assay (TMSB10) were run in triplicate using real-time PCR mixes from multiple suppliers on another trusted real-time PCR system (Figure 1). A six-log dilution series was performed starting from 1000 ng cDNA input. TaqMan Fast Advanced Master Mix achieved efficiencies within the optimal range for both ACTB (98%) and TMSB10 (99%), demonstrating reliable performance for duplex analysis.

TaqMan Fast Advanced Master Mix demonstrates enhanced sensitivity for low-level target detection
When assessing reaction performance, a lower Cq value can be associated with higher sensitivity, as it suggests that small amounts of the target sequence can be detected earlier in the amplification process. A FAM-labeled MADC6 assay was run in triplicate using real-time PCR mixes from multiple suppliers on a real-time PCR instrument from another global qPCR provider (Figure 2). A six-log dilution series was performed starting from 1000 ng cDNA input. The TaqMan Fast Advanced Master Mix detected the target at an earlier cycle for each dilution, indicating enhanced sensitivity for low-level target detection.

TaqMan Fast Advanced Master Mix facilitates reliable amplification in the presence of PCR inhibitors
Maintaining consistent amplification even with high levels of common PCR inhibitors helps ensure accurate results under challenging sample conditions. To assess tolerance, a TaqMan GAPDH assay was performed with high concentrations of hemoglobin, hematin, heparin, and humic acid spiked into 1 ng of cDNA per 10‑µL reaction (Figure 3). Reactions were run in triplicate using several different commercially available qPCR master mixes using a qPCR system from another global instrument manufacturer. A no-template control was included but not shown. TaqMan Fast Advanced Master Mix showed greater tolerance than competitor mixes, which were completely inhibited in three of the four conditions.

When monitoring SYBR Green assay specificity using melt curve analysis, a single peak suggests amplification of a single product and thereby target specificity. Real-time PCR was performed using universal human reference (UHR) cDNA input of 1 ng/reaction and primers targeting TLR2, SO1a, AO4a, and S11a (Figure 4). Reactions were run in triplicate using real-time PCR mixes from multiple suppliers on another leading real-time PCR system. PowerTrack and PowerUp SYBR Green master mix demonstrates single melt curve peaks for assays that demonstrated non-specific signal using other reagents, suggesting enhanced specificity.
