Applied Biosystems PrepSEQ chemistry is a universal sample preparation workflow for contaminant and impurity testing designed to recover and purify nucleic acids from complex bioprocessing samples. Compatible with multiple applications, PrepSEQ chemistry supports workflows including:

  • Residual host cell DNA quantitation
  • Mycoplasma detection
  • Minute Mouse Virus (MMV) detection
  • Viral contaminant detection

Using magnetic bead-based chemistry, PrepSEQ kits efficiently isolates nucleic acids while helping remove proteins, salts, buffers, and other process-related contaminants that may interfere with downstream analysis. The purified DNA or RNA can then be used in qPCR and other molecular testing workflows.

How PrepSEQ Works

PrepSEQ chemistry uses magnetic bead-based purification to isolate nucleic acids from complex bioprocessing samples. The workflow follows six simple steps that can be performed manually or automated depending on your laboratory's throughput requirements.

Samples are lysed to release nucleic acids from cells, viral particles, or other biological material, creating a total lysate for purification.

Magnetic particles and a proprietary binding solution are added to the lysate, allowing DNA or RNA to selectively bind to the magnetic beads while unwanted sample components remain in solution.

The nucleic acid-bound magnetic particles provide an efficient mechanism for separating the target DNA or RNA from complex sample matrices while maintaining high recovery.

A series of purification washes removes proteins, salts, inhibitors, and other process-related contaminants, helping improve nucleic acid purity for downstream molecular analysis.

A magnetic field captures the nucleic acid-bound particles while contaminants are removed, simplifying sample cleanup and minimizing sample loss.

Purified nucleic acids are released from the magnetic particles into an optimized elution buffer, producing a PCR-compatible nucleic acid solution ready for downstream analysis.

The purified DNA or RNA is suitable for a variety of downstream contaminant and impurity testing applications, including:
