CultureOne Supplement enhances the differentiation of neural stem cells (NSCs) to neurons and can be used with primary rodent neurons or psc-derived neurons. A superior alternative to anti-mitotic agents like Ara-C or mitomycin C, CultureOne Supplement not only eliminates more than 75% of contaminating neural progenitor cells with minimal cell death, it also accelerates neuronal maturation. Using CultureOne Supplement in neuronal differentiation medium for two weeks yields more mature, functional neurons, reduces cell clumping, and allows maintenance of robust cultures for over a month.

CultureOne Supplement is a chemically-defined, serum-free supplement, provided as a 100X liquid reagent that can be easily added to any conventional neuronal differentiation medium. It should be used in place of other enrichment approaches such as Ara-C, mitomycin-C, fluorouracil (5-FU), or DAPT treatment.

Improved downstream assays

Analysis of NSC-derived neurons and primary rodent neurons can be difficult due to the presence of overgrown progenitor cells in the culture. These invasive cells complicate imaging and electrophysiology experiments and can affect the physiological relevance of gene expression assays. CultureOne Supplement efficiently controls the proliferation of both neural progenitor cells and glial cells, resulting in neurons of the highest purity and accelerating maturation and function.

Non-toxic proliferation control

Traditional methods to control proliferating cell populations involve treatments with anti-mitotic agents like cytosine arabinoside (Ara-C), which can be toxic to neurons. CultureOne Supplement controls proliferation without cytotoxicity. Cultures treated with CultureOne Supplement show better neurite outgrowth, less cell clumping, and reduced cell death compared to those treated with Ara-C or mitomycin C.

Accelerated maturation of differentiated neurons

NSC-derived neurons and primary rodent neurons treated with CultureOne Supplement express mature physiological markers faster and develop longer neurites than untreated cultures. These neurons also have significantly higher numbers of voltage-gated calcium ion channels, important for neural maturity and excitability.

Extended culture duration

CultureOne Supplement enables maintenance of enriched, differentiated neurons for five weeks or longer, unlike conventional methods that result in proliferating progenitor cells, causing clumping and peeling off the plate.

What are the storage conditions for CultureOne Supplement? Can I store it at 4 degrees C and for how long? Can I aliquot unused product and freeze it at -20 degrees C?

We recommend that you thaw the CultureOne Supplement and then aliquot it out for working stocks and re-freeze the aliquots. We have found that the product can undergo up to 3 freeze-thaws without loss of activity. CultureOne-supplemented media is good for 2 weeks stored at 4 degrees C in the dark.

I am using the B-27 Plus Neuronal Culture System. What do I do if my neuronal cells are not attaching or are clumped?

In primary neuronal cultures, this is typically a function of uneven coating or if neurons are plated at too high of a density. In NSC differentiation experiments, this could be due to overproliferation of neural progenitor cells. Differentiating with the B-27 Plus Neuronal Culture System and CultureOne Supplement can eliminate more than 75% of neural progenitor cells and the clumps they form. I am planning to use the B-27 Plus Neuronal Culture System. How can I control the number of glial cells in my neuronal culture?

Add CultureOne Supplement at day 0 with the B-27 Plus Neuronal Culture System to fully suppress both astrocytes and oligodendrocytes with no detrimental effect on neurons. Delaying the addition of CultureOne Supplement to later time points results in increasing levels of astrocytes.
