Thermo Scientific Pierce EMCS is an amine-to-sulfhydryl crosslinker that contains NHS-ester and maleimide reactive groups at opposite ends of a medium-length spacer arm (9.4 angstroms).
Features of EMCS:
• Reactive groups: NHS ester and maleimide • Reactive towards: amino and sulfhydryl groups • C6-homolog of aliphatic spacer series which includes AMAS, BMPS, GMBS and EMCS • NHS ester end couples with primary amines at pH 7-9 to form stable amide bonds • Maleimides react with -SH groups at pH 6.5-7.5, forming stable thioether linkages • Non-cleavable • Water-insoluble (dissolve first in DMF or DMSO); compare to Sulfo-EMCS • Increased sphere of coupling vs. GMBS • Short aliphatic spacer has low potential for eliciting an immune response

Mechanism of reversal of phospholamban inhibition of the cardiac Ca2+-ATPase by protein kinase A and by anti-phospholamban monoclonal antibody 2D12.
Authors:Chen Z, Akin BL, Jones LR
Journal:J Biol Chem
PubMed ID:17548345
'Our model of phospholamban (PLB) regulation of the cardiac Ca(2+)-ATPase in sarcoplasmic reticulum (SERCA2a) states that PLB binds to the Ca(2+)-free, E2 conformation of SERCA2a and blocks it from transitioning from E2 to E1, the Ca(2+)-bound state. PLB and Ca(2+) binding to SERCA2a are mutually exclusive, and PLB inhibition of ... More
Miniature single-particle immunoassay for prostate-specific antigen in serum using recombinant Fab fragments.
Authors:Härmä H, Tarkkinen P, Soukka T, Lövgren T
Journal:Clin Chem
PubMed ID:11067810
Quantitative, miniaturized nucleic acid assays and immunoassays can be developed with single microparticles, microfluorometric detection, and intrinsically fluorescent lanthanide chelates in a multiple assay format to decrease reagent consumption, cost, and assay time. We used recombinant Fab fragments to capture and detect free and total prostate-specific antigen (PSA) from serum ... More
Characterizing phospholamban to sarco(endo)plasmic reticulum Ca2+-ATPase 2a (SERCA2a) protein binding interactions in human cardiac sarcoplasmic reticulum vesicles using chemical cross-linking.
Authors:Akin BL, Jones LR
Journal:J Biol Chem
PubMed ID:22247554
Chemical cross-linking was used to study protein binding interactions between native phospholamban (PLB) and SERCA2a in sarcoplasmic reticulum (SR) vesicles prepared from normal and failed human hearts. Lys(27) of PLB was cross-linked to the Ca(2+) pump at the cytoplasmic extension of M4 (at or near Lys(328)) with the homobifunctional cross-linker, ... More
PNA microarrays for hybridisation of unlabelled DNA samples.
Authors:Brandt O, Feldner J, Stephan A, Schröder M, Schnölzer M, Arlinghaus HF, Hoheisel JD, Jacob A
Journal:Nucleic Acids Res
PubMed ID:14500847
Several strategies have been developed for the production of peptide nucleic acid (PNA) microarrays by parallel probe synthesis and selective coupling of full-length molecules. Such microarrays were used for direct detection of the hybridisation of unlabelled DNA by time-of-flight secondary ion mass spectrometry. PNAs were synthesised by an automated process ... More
Role of leucine 31 of phospholamban in structural and functional interactions with the Ca2+ pump of cardiac sarcoplasmic reticulum.
Authors:Chen Z, Stokes DL, Jones LR
Journal:J Biol Chem
PubMed ID:15644311
The ability of two loss-of-function mutants, L31A and L31C, of phospholamban (PLB) to bind to and inhibit the Ca(2+) pump of cardiac sarcoplasmic reticulum (SERCA2a) was investigated using a molecular cross-linking approach. Leu(31) of PLB, located at the cytoplasmic membrane boundary, is a critical amino acid shown previously to be ... More
