Beyond dioxins - Obtaining accurate compound identification in environmental samples with GC Orbitrap MS

In this presentation hear from Dr. Kjell Hope from Pacific Rim Labs on their approach to non-targeted screening and quantitation using GC Orbitrap MS. Environmental samples were extracted with the aim of preserving sample integrity whilst sufficiently cleaning the sample of background matrix. Extract analysis was performed using high-resolution accurate mass GC Orbitrap MS in full scan mode at 60,000 resolution. Analysis in full scan (untargeted) allows for a complete chemical profile to be collected from the sample. This data was then processed to screen for an unlimited number of compounds, thereby increasing scope of analysis beyond what is possible using targeted methods such as GC-MS/MS or GC Magnetic Sector instrumentation. In this study, samples were processed and compared using Thermo Scientific Compound Discoverer Software to identify features of interest. These suspect features were then screened against NIST 2023 and a high-resolution accurate mass environmental contaminants library. The complete workflow, which aims for fast and accurate compound identification, will be discussed.

About the speakers

Kjell Hope

Kjell Hope has been working in chemical analysis for 8 years. He obtained a master’s degree in chemistry from Orebrö University in Sweden analysing PFAS in water collected off the east coast of Africa. He has contributed to many projects in private industry and academia, including the UNEP global monitoring of persistent organic pollutants, project Campania Trasparente, screening PCBs in various foodstuffs, and ongoing MS/MS method development at Pacific Rim Labs.

Heinz Mehlmann

Dr. Heinz Mehlmann is a Senior Product Specialist for the GC-HRMS. His background is in Electronics, and he has a Ph.D. in Organic Chemistry. He started at Thermo Fisher Scientific as a Global Support Specialist and was involved in the development of the DFS magnetic Sector GC-HRMS. He has 20 years of experience in high resolution mass spectroscopy particularly in the field of Dioxin and POPs analysis.

Nicolas Warner

Nicholas is a product specialist with a strong scientific background in GC and GC-MS applications. Originally from Canada, he has worked as an environmental researcher for 14 years within Norway before moving to join the Thermo Fisher Scientific team in Bremen, Germany in 2021. With a background in Environmental/Analytical chemistry, he is a specialist in the analysis of emerging harmful substances in the environment using gas chromatography mass spectrometry and high-resolution mass spectrometry.

Breaking Down POPs & PFAS : Sample to Solution looking at sample preparation and PFAS analysis, specifically untargeted analysis

New advancements have been made which allows for a more efficient and controlled extraction of persistent organic pollutants (POPs), using parallel gas assisted accelerated solvent extraction, in-line cleanup and automated solvent concentration in the same platform. We present here performance data from soil samples for different POPs. Unlike traditional methods such as liquid-solvent extraction followed by N2 stream evaporation, the fully automated solvent extraction, in-line cleanup and evaporation system saves time, solvent, and labor, while ensuring high reproducibility and productivity for analytical testing.

One of the main emerging POPs of concern is PFAS (Per- and Polyfluoroalkyl Substances) and the vast amount and variety of PFAS compounds having different chemical properties creates analysis challenges and opportunities to apply multiple techniques like GC- and LC-MS for the analysis. To screen and/or quantitate the relevant PFAS compounds appropriate sample preparation, chromatography and instrument settings, and experiments are required due to the nature of different matrices. We will present how to analyze large number of compounds utilizing full scan with high resolution and has the advantage to be able to review data retrospectively.
