Compare TaqMan Fast Advanced Master Mix to Your Probe-Based Supermix
Low-copy targets can be particularly challenging in multiplex qPCR, where analytical sensitivity, precision, and PCR efficiency can all influence reliable quantification. In comparative testing on a Bio-Rad™ CFX Opus™ Real-Time PCR System, TaqMan Fast Advanced Master Mix was evaluated against Bio-Rad™ SsoAdvanced™ Universal Probes Supermix, Bio-Rad™ iTaq™ Universal Probes Supermix, and Bio-Rad™ iQ™ Multiplex Powermix. Under the study conditions, TaqMan Fast Advanced Master Mix demonstrated greater or equal precision across the evaluated multiplex targets compared with the Bio-Rad supermixes tested. At 0.0256 ng cDNA per reaction, replicate amplification profiles were evaluated across FAM, VIC, and JUN targets. Across the dilution series, TaqMan Fast Advanced Master Mix demonstrated PCR efficiencies of 101%, 102%, and 97% for FAM, VIC, and JUN, respectively.

Melt curve analysis can help assess SYBR Green assay specificity. A single melt peak suggests amplification of a single product, while additional peaks can indicate nonspecific amplification or primer-dimer formation.
In comparative testing on a Bio-Rad™ CFX Opus™ Real-Time PCR System, melt curve profiles were evaluated across three representative SYBR Green assays using PowerTrack and PowerUp SYBR Green Master Mixes and two Bio-Rad supermixes.
